Abstract

For millennia, human culture framed the desire to transcend aging within mythic, theological, and literary archetypes, ranging from the Epic of Gilgamesh to the Faustian bargain. In contemporary medicine, this existential pursuit has shifted to molecular laboratories, where the discipline of geroscience seeks to intervene in fundamental biological mechanisms of decay. Metformin a widely prescribed, low-cost biguanide historically limited to managing type 2 diabetes mellitus has recently emerged as a primary artifact in this scientific transition. Operating at the boundary between mundane clinical therapeutics and anti-aging bio-transcendence, metformin’s repositioning as a geroprotective candidate offers a critical focal point for humanistic inquiry. This paper critically evaluates the biological claims surrounding metformin including AMP-activated protein kinase (AMPK) activation, mechanistic target of rapamycin (mTOR) inhibition, autophagic degradation, chromatin fragment dynamics, and epigenetic biological clocks by embedding them within theoretical frameworks of Medical Humanities, Philosophy of Science, and Bioethics. We analyze how biomedical discourses construct aging either as a natural teleological process or as a treatable pathology, examine the ethical implications of non-human primate research (such as the 40-month cynomolgus monkey trial), and deconstruct the socio-economic paradigm shift proposed by the Targeting Aging with Metformin (TAME) trial framework. By synthesizing empirical literature with critical theory, this study demonstrates how geroprotective science reflects evolving cultural anxieties surrounding mortality, social utility, and the ethics of bodily extension.
